4 studs and if you drew a circle which passed the centre of each stud hole it would have 100mm diametre

I thought it was if you measure the distance between centres of two opposite studs you would get 100mm

In answer to your first question (Which was sort of answered by the first reply)..
4 x 100
the first number defines a number of points (holes) and the second number defines the diameter of an imaginary cirlce that passes through the center of all of the holes.
So 4 x 100 means there are four holes and the center of opposite holes is 100mm apart so if you put the wheel with one bolt hole at top, one at the right, one at the bottom and one on the left the distance between top and bottom will be 100mm and so will the distance between left and right.
