Aim: To understand the molecular basis of the action of methylated chromatin binding proteins through knowledge of their three-dimensional structure and analysis of their interactions with other components of chromatin.

Objectives:

1/ To discover the three dimensional structure of a methylated chromatin binding protein by NMR spectroscopy.
2/ To discover how the protein interacts with other chromatin components.

Background: Methylated chromatin binding proteins such as MBD and chromo-domain proteins bind to covalently modified nucleic acids or protein components of chromatin to regulate chromatin structure and influence gene transcription. These interactions are the basis of epigenetic phenomena that are key to the stably inherited changes in gene transcription that underlie cellular differentiation and other epigenetic phenomena. Defects in the regulation and interpretation of the methylation signals are implicated in many disease states including cancer, and in ageing.

Approach: The target methylated chromatin binding protein will be expressed in E.coli and purified for structural studies using high field NMR spectroscopy. Functional studies will be carried out using a variety of biophysical techniques (calorimetry, Biacore, analytical ultracentrifugation) to investigate the interactions between the protein and other proteins or nucleic acids. The structural information will be used to inform biological experiments.

Methods Training: The project will utilise multidimensional NMR spectroscopy to determine the three-dimensional structures of the proteins and to study their complexes with ligands. It will utilise a recently acquired 600MHz NMR spectrometer equipped with a cryoprobe. Computer modelling, biophysical techniques, molecular biology and recombinant protein technologies will be used to exploit the structural data. All research students in IBLS also participate in compulsory and elective elements of our postgraduate training programme, which includes a wide range of specific and generic (transferable) skills and experience.
